Selene and Eve are pursued by Antigen, which is led by Dr. Jacob Lane, a Lycan in disguise. Lane wants Eve's DNA to create a "Super-Lycan" that will be immune to silver and powerful enough to destroy the Vampires. Selene is aided by David, a young Vampire who believes that the Vampires should fight back against the humans and Lycans. Together, they must protect Eve and find a way to stop Antigen's plans.

The film begins with a recap of the events of the previous films, showing how the humans discovered the existence of the Vampires and Lycans and began a "Purge" to exterminate them. Selene and her lover Michael Corvin are captured during the Purge. Selene is frozen in cryogenic suspension at Antigen, a biotech corporation. Twelve years later, she is awakened by a young girl named Eve, who is revealed to be her daughter with Michael.
